In addition to being animal and human pathogens, they have been isolated from soil, plant material, waste water, and dairy products. C.diphtheriae produces the potent diphtheria A-B exotoxin, which is usually encoded by a bacteriophage or by an integrated prophage as in this strain. The toxin ADP-ribosylates elongation factor EF-2, inhibiting protein biosynthesis and killing the host cells. Expression of the toxin is under control of the chromosomal iron-sensing regulator DtxR, and transcription is is induced under low iron concentrations. Diphtheria is an acute disease that usually affects the tonsils, throat, nose and/or skin. It is passed from person to person by droplet transmission, usually by breathing in diphtheria bacteria after an infected person has coughed or sneezed. Classical diphtheria can lead to breathing problems, heart failure, paralysis and sometimes death. Endocarditis and (quite rarely) pneumonia can also be caused by C.diphtheriae. Vaccines are based on inactivated toxin. Strain CDCE 8392 (CDC-E8392) was isolated from a diphtheria patient and was originally from the Centers for Disease Control and Prevention. The strain is A-B exotoxin positive. Comparison of 13 fully sequenced C.diphtheriae genomes shows the core genome to be about 1632 genes, with a (May 2012) 4786 gene open pangenome. Approximately 65 unique genes are expected on each newly sequenced genome, mainly found in genomic islands that seem to be due to horizontal gene transfer. Each sequenced strain encodes at least 2 pilus gene clusters, indicating the mechanisms of host cell adherence may be quite diverse (adapted from PMID 22505676)."xsd:string
